This study aimed to determine the antiulcerogenic and antioxidant activities of Psyllium (Plantago ovata Forssk) seed ethanolic extract in rats. We assessed the antioxidant potential using free radical scavenging on DPPH, ß-carotene bleaching activity, ferric reducing power, and hydroxyl radical scavenging activity. In the antiulcerogenic study, pre-treatment with Plantago ovata seeds ethanolic extract (POE) (400 mg/kg b.wt) significantly protected against ethanol-induced gastric ulcer in rats by decreasing the ulcer index value and preserving the integrity of the gastric mucosa. The oxidative stress status in the stomach tissues showed a significant increase in the antioxidant enzyme levels of superoxide dismutase, catalase, and glutathione peroxidase with a significant decrease in lipid peroxidation during pre-treatment with POE. In conclusion, the POE protects against gastric ulcer due to its antioxidant potential and presence of bioactive molecules.


O presente estudo teve como objetivo determinar as atividades antiulcerogênica e antioxidante das sementes de Psyllium (Plantago ovata Forssk) em ratos. O potencial antioxidante foi avaliado utilizando o método do sequestro do radical livre DPPH, autooxidação do ß-caroteno, poder redutor de ferro e atividade de sequestro do radical hidroxila. No estudo antiulcerogênico, o pré-tratamento com o extrato etanólico das sementes de Plantago ovata (POE) (400 mg/Kg b.wt) reduziu a úlcera gástrica induzida pelo etanol em ratos, diminuindo o valor do índice de úlcera e preservando a integridade da mucosa gástrica. O estudo do estresse oxidativo nos tecidos estomacais mostrou um aumento significativo dos níveis das enzimas antioxidantes superóxido dismutase, catalase e glutationa peroxidase, com uma diminuição significativa da peroxidação lipídica enquanto pré-tratamento com POE. Em conclusão, o POE protege contra úlcera gástrica devido aos seus potenciais antioxidantes e à presença de moléculas bioativas.

Sturgeon cartilage, which is rich in chondroitin sulfate (CS), is usually discarded during sturgeon utilization. In this paper, CS was isolated from large hybrid sturgeon skull and backbone and named SCS and BCS, respectively. Their structures were investigated via Fourier transform infrared (FT-IR) spectroscopy, nuclear magnetic resonance (NMR) spectroscopy and high performance liquid chromatography (HPLC). The average molecular weights of SCS and BCS were ~ 30-44 kDa. Disaccharide analysis indicated that SCS and BCS had similar chemical structures and were composed of ΔUA-[1 â†’ 3]-GalNAc (ΔDi0S, 14.71%, 16.04%), ΔUA-[1 â†’ 3]-GalNAc-4 s (ΔDi4S, 32.01%, 37.78%) and ΔUA-[1 â†’ 3]-GalNAc-6 s (ΔDi6S, 53.27%, 46.18%). The gastroprotective effect of SCS and BCS were studied using a rat model of ethanol-induced gastric ulcers. Both SCS and BCS had apparent gastroprotective activity and their ulcer inhibition rate reached ~ 35%-45%, which was similar to that of omeprazole (~41%). These results provide useful strategies for the high-value utilization of sturgeon cartilage.

Phy-Blica-D is a traditional Thai polyherbal formula that has reduced oxidative stress in non-communicable diseases. However, evidence supporting the gastroprotective effects of Phy-Blica-D has not been previously reported. Therefore, this study aimed to evaluate the gastroprotective effects of Phy-Blica-D against gastric ulcers in rats and investigate the potential underlying mechanism. To estimate the possible mechanisms of action, we examined the levels of oxidative stress markers, such as reactive oxygen species (ROS) and malondialdehyde (MDA), as well as antioxidant enzymes, including catalase (CAT), superoxide dismutase (SOD), and glutathione (GSH). According to our results, rats treated with only 80% ethanol (vehicle group) exhibited significant increases in their ulcer area and ulcer index (UI). Moreover, the levels of ROS and MDA markedly increased in the vehicle group compared with the normal control group. Daily oral administration of Phy-Blica-D (500 and 1000 mg/kg) for 7 days not only significantly decreased the ulcer area and UI, but also remarkably decreased the ROS and MDA levels in gastric tissue. Gastric ulcers induced by ethanol had significantly decreased antioxidant enzyme activities (CAT and SOD) and non-enzymatic antioxidant (GSH), whereas pretreatment with Phy-Blica-D significantly improved the activities of CAT, SOD, and GSH. Moreover, after exposure to ethanol, the rats exhibited a significantly increased level of inducible nitric oxide synthase (iNOS), which was reduced after treatment with Phy-Blica-D. These findings suggest that Phy-Blica-D potentially exerts its gastroprotective effects by suppressing oxidative stress and stimulating antioxidant enzymes, which is one of the causes of destruction of cell membranes, and it is involved in the pathogenesis of acute gastric ulcers induced by ethanol.

A variety of bioactivities has been established for fucoidan extracted from sea cucumber, whereas its structure-activity relationships have been seldom investigated. In this study, sea cucumber ( Thelenota ananas) fucoidans with different molecular weights were prepared with enzymatic degradation. The chain stiffness and molecular size decreased with the decreasing of molecular weight. Fucoidans with molecular weight of 1380.0, 828.7, and 483.0 kDa exhibited random coil conformations, while fucoidan molecular weight of 215.0 kDa existed as sphere in solution. All examined fucoidans could effectively prevent the ethanol-induced gastric ulcer, of which mechanism involved antioxidation and anti-inflammation. Within the range of the study, the performance of fucoidans adopted random coil conformations declined with the decreasing of molecular weight, and the performance recovered when the chain conformation transited from coil to sphere, indicating the subtle influences of molecular weight and chain conformation on the gastric protective activity of sea cucumber fucoidan.

Aronia melanocarpa (Michx.) Ell. belongs to the Rosaceae family. The purpose of this study is to explore the gastroprotective effect of the Aronia melanocarpa hydro-alcoholic extract (AMHAE) against ethanol-induced gastric ulcer in a rat model. Different concentrations (50, 100, and 200 mg/kg) of AMHAE, or 30 mg/kg of omeprazole, significantly inhibited the gastric injury formation. The ethanol-induced ulcer group showed significant increases of malondialdehyde (MDA), myeloperoxidase (MPO), tumor necrosis factor (TNF)-α, nuclear factor-kappaB p65 (NF-κB p65), and monocyte chemoattractant protein (MCP)-1, and decreased activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-px), and interleukin (IL)-4. However, AMHAE (200 mg/kg) pretreatment significantly reversed the altered pathophysiological levels of these biomolecules to near normal stages. The gastroprotective activity of AMHAE was abolished by pretreatment with l-NAME, naloxone, capsazepine, and indomethacin, demonstrating the participation of nitric oxide (NO), opioids, TRPV (vanilloid receptor-related transient receptor potential), and prostaglandins in AMHAE-assisted gastroprotection against ethanol-induced gastric injuries. This gastroprotective effect of AMHAE might be due to the downregulation of TNF-α-based NF-κB, MCP-1 signaling and strong antioxidant properties.

Bilobalide (BI) has been widely known as a unique constituent extracted from Ginkgo biloba. The aim of the current study was to reveal the potential efficacy as well as the underlying mechanism of the action of BI on ethanol-induced lesion in gastric mucosa in vivo/vitro. Ethanol (0.2ml/kg) was applied to induce gastric ulcer mice model. Our results indicated that treatment with BI markedly decreased the levels of interleukin-6 (IL-6), IL-1ß and tumor necrosis factor-α (TNF-α) in vivo. Additionally, BI intervation exhibited elevated myeloperoxidase (MPO) level in stomach, increased superoxide dismutase (SOD) activity and decreased malonaldehyde (MDA) content in serum and stomach when compared with those of the model group. It could be also observed that inhibited MAPK/NF-κB pathway expressions occurred after BI treatment both in vivo and in vitro. Taken together, BI exerted a gastro-protective effect against gastric ulceration, which was presumed to be associated with MAPK/NF-κB pathway.

An extracellular polysaccharide of Lachnum sp. (LEP) was purified by DEAE-cellulose 52 column chromatography and Sepharose CL-6B column chromatography. LEP-2a was identified to be a homogeneous component with an average molecular weight of 3.22 × 10(4)Da. The structure of LEP-2a was characterized by chemical and spectroscopic methods, including methylation analysis, periodate oxidation-smith degradation, infrared spectroscopy and NMR analysis. Results indicated that LEP-2a was a (1→3)-,(1→6)-ß-D-Glcp, whose branch chain was consist of two d-glucopyranosyl residues linked by ß-1,3-glycosidic linkage, which was linked at C6 of the backbone chain by ß-1,6-glycosidic linkage. To study the protective effects of LEP-2a on the ethanol-induced gastric ulcer in mice, LEP-2a (100, 200 and 400mg/kg/d) was given to mice by gavage for 2 weeks. Results showed that LEP-2a significantly decreased the ulcer bleeding areas, pepsin activity, gastric juice volume, gastric juice total acidity and the malondialdehyde (MDA) content in serum. Meanwhile, the superoxide dismutase (SOD) increased significantly. The above findings suggested that LEP-2a had a significant preventive effect against the ethanol-induced gastric ulcer.

Epoxy-carvone is a monoterpene present in essential oils of various plants. It is a derivative of carvone, which has an epoxy group instead of the α- and β-unsaturated ketone group present in carvone. As recent studies have shown that several alcohol terpenes and compounds containing α, β-unsaturated ketone groups present antiulcer effect, the main of the present study was to evaluate the antiulcer effect of epoxy-carvone. The models of ulcers induced by ethanol and indomethacin were used in this study. Epoxy-carvone at the dose of 1 mg/kg did not present antiulcer effect against ulcer induced by ethanol, but at the doses of 10, 30 and 50 mg/kg it presented gastroprotective effect in both ulcer models. Epoxy-carvone also did not affect the gastric secretion in the pylorus ligation test. Moreover, pretreatment with indomethacin or L-nitroarginine methyl ester did not reverse the gastroprotection produced by this monoterpene. This study showed that epoxy-carvone presents antiulcer effect and suggests that this effect does not involve either antisecretory activity or increase of the nitric oxide and prostaglandin synthesis.
